Epithelial Functions and Smooth Muscle Contraction in Regards to Inositol-6-Phosphate Degradation in Laying Hens

The present doctoral thesis evaluated the physiological functions of the laying hen gut regarding epithelial and smooth muscle functions. The specific focus was set on the myo-inositol (MI) formation and transport. The potential activity of InsP6 degradation by bacterial phytases and phosphatases was studied. The mucosal transport of the end product of InsP6 degradation, myo-Inositol, was investigated for the first time in chickens.
Experiments were done using laying hens from two commercial lines (Lohmann LSL and LB). Hens were euthanized and the intestine of jejunum, ileum and the caeca, as well as the digesta of the small intestine and the caeca was used. Mucosal barrier function of the ileum and jejunum and MI transport was measured in Ussing chambers. Changes of the short-circuit current were measured after MI addition in the mucosal chamber with concurrent determination of the transepithelial resistance of the tissue. Further, the amount of MI in the mucosal and serosal buffer, as well as in the mucosal samples was analyzed. Intestinal smooth muscle from the caeca of the same animals was used to measure the contraction of caecal circular smooth muscle strips by an isometric force transducer determining the frequency, height and intensity. Near the physiological contraction, that was used as control, the reaction to different drugs and buffers was tested effecting smooth muscle contraction at different levels. Finally, outer membrane vesicles were isolated from the digesta samples of the duodenum, jejunum, ileum and the caeca of the hens. Phytase and phosphatase activity of the remained digesta samples and the OMV samples was measured spectrophotometrically using a modified kit for phytase and phosphatase activities. Furthermore, proteomic analyzes of digesta and OMV pool samples from duodenum, jejunum, ileum and caeca were performed.
The hypothesis that MI is transported by an active ion-driven transport system could not be approved in Ussing chamber experiments. MI was not detectable in the serosal buffer, nor in the lysates of mucosal tissue cytoplasm. This leads to the conclusion that transepithelial MI transport process in chickens is more complex than it was presumed and is probably influenced by still unknown regulations or metabolic processes lacking in vitro. Referring to this, the direct inclusion of MI into the cell membrane by PIP-synthesis is imaginable. Isometric force measurements provided the first key dates to characterize the contraction of the caecal smooth muscle in chicken. The contraction behavior of caecal smooth muscles in laying hens was similar to other species. Moreover, differences between the two laying lines concerning the response to carbachol were indicated leading to the assumption that there are maybe further differences between the two lines regarding their intestinal physiology. The analyses of the digesta and bacterial OMVs according their capacity to catalyze InsP6 confirmed the activity of phytases and phosphatases in all intestinal sections. The activity of both enzymes was higher in the digesta samples compared to the OMV samples. Unfortunately, there are still open questions regarding the influence of free phosphorus in the samples during the measurement. However, proteome data of OMV pool samples confirmed the hypothesis that bacterial phytases and phosphatases packed in OMVs are present in the intestinal tract of chickens.
The present data provides more knowledge and improves the understanding of the gut physiology and the phytate degradation in the laying hen. These findings are the basis for future research in this important field and can support the establishment of new and modern conceptions in the animal production including a high animal welfare and protection of the environment.
 
Die vorliegende Doktorarbeit untersuchte die physiologische Funktion des Darms der Legehenne in Bezug auf die Funktion des Epithels und der glatten Muskulatur. Dabei lag der Fokus auf der Entstehung und dem Transport von Myo-Inositol (MI). Der potentielle Abbau von InsP6 durch bakterielle Phytasen und Phosphatasen wurde untersucht. Zum ersten Mal wurde der mukosale Transport des Endproduktes des InsP6 Abbaus, Myo-Inositol, in Hühnern erforscht.
Um dieser Forschung nachzugehen wurden Legehennen von zwei verschiedenen Linien verwendet. Die Hennen wurden eingeschläfert und der Darm von Jejunum, Ileum und den Caeca, sowie der Darminhalt vom Dünndarm und den Caeca wurden verwendet. Die Integrität der Mukosa im Ileum und Jejunum und der Transport von MI wurden in Ussing Kammern untersucht. Neben einer konstanten Ermittlung des transepithelialen Widerstands, wurden Veränderungen im Kurzschlussstrom gemessen, nachdem MI in der mukosalen Kammer zugegeben wurde. Zudem wurde die Menge an MI im mukosalen und serosalen Puffer, sowie in den Mukosa Proben, ermittelt. Die glatte Muskulatur der Blinddärme derselben Tiere wurde verwendet, um die Kontraktion von Muskel Streifen des zirkulären glatten Muskels im Caecum mit Hilfe eines Kraftaufnehmers zu messen. Dabei wurden die Frequenz, maximale Höhe und Intensität der Kontraktionen ermittelt. Neben der physiologischen Kontraktion, die als Kontrolle fungierte, wurde auch die Reaktion auf verschiedene Puffer und Substanzen getestet, die die Kontraktion der glatten Muskulatur auf verschiedenen Ebenen beeinflussen. Zuletzt folgte die Isolation der Vesikel der äußeren Membran aus dem Darminhalt des Duodenums, Jejunums, Ileums und der Caeca der Hennen. Die Phytase und Phosphatase Aktivität der OMV Proben und des übrigen Darminhalts wurde spektrophotometrisch mit einem modifizierten Kit für die Phytase und Phosphatase Aktivität gemessen. Zudem wurden Proteom Analysen von Pool Proben der Digesta und der OMVs von Duodenum, Jejunum, Ileum und Caecum durchgeführt.
Die Hypothese, dass MI über ein elektrogenes ionen-getriebenes Transportsystem aufgenommen wird, konnte in den Ussing Kammer Versuchen nicht bestätigt werden. MI war weder im serosalen Puffer, noch in den Lysaten des mukosalen Gewebes nachweisbar. Das führt zu dem Schluss, dass der MI Transport im Huhn wesentlich komplizierter ist als zunächst angenommen und dass er möglicherweise durch bisher unbekannte Regulationsmechanismen oder metabolische Prozesse, die in vitro fehlen, beeinflusst wird. Im Hinblick darauf ist ein direkter Einbau von MI in die Zellmembran über die PIP Synthese denkbar. Die isometrische Kraftmessung liefert die ersten Daten über die Kontraktion der glatten Muskulatur im Caecum des Huhns. Ein ähnliches Kontraktionsverhalten zu anderen Spezies wurde in der glatten Muskulatur des Caecums beobachtet. Zudem wurden Unterschiede zwischen den beiden Linien bezüglich ihrer Reaktion auf Carbachol festgestellt, was zu der Annahme führt, dass noch mehr Unterschiede in der intestinalen Physiologie der beiden Linien existieren. Die Untersuchung der Digesta und der OMV Proben in Bezug auf ihre Fähigkeit InsP6 abzubauen zeigte eine Phytase- und Phosphataseaktivität in allen Darmabschnitten. Die Aktivität beider Enzyme war in den Digesta Proben höher als in den OMV Proben. Der Einfluss von freiem, in den Proben enthaltendem Phosphor auf die Messungen konnte bedauerlicherweise nicht geklärt werden. Jedoch untermauern die Proteom Messdaten der OMV Pool Proben die Hypothese, dass in OMVs verpackte, bakterielle Phytasen und Phosphatasen im Verdauungstrakt des Huhns vorhanden sind.
Zusammenfassend tragen die hier gezeigten Daten zu einem besseren Verständnis für die Darmphysiologie und den Phytatabbau in der Legehenne bei. Die Ergebnisse schaffen eine Basis für die weitere Forschung auf diesem wichtigen Gebiet und können zur Entwicklung neuer und moderner Konzepte in der Tierhaltung beitragen, die einen hohen Standard in Tierwohl und Umweltschutz beinhalten.
